Capillary Isotachophoresis Determination of Trace Oxidized Glutathione in Blood

Abstract: A capillary isotachophoresis (CITP) method performed in a column-coupling apparatus has been developed for the simultaneous determination of glutathione (GSH) and glutathione disulfide (GSSG) concentrations in blood samples. The determination of GSSG and GSH concentrations in biological samples is important because of their roles in oxidative stress. Different concentrations of a leading ion in the coupled columns (concentration cascade) and a large volume (37 µ ) of the injected sample facilitated a GSSG conc… Show more

“…This may be the cause of the higher values in comparison to ours. Bodor et al (2018) used two different methods for the evaluation of glutathione in the whole blood of dairy cows. The enzymatic spectrophotometric method and isotachophoresis were applied to compare the concentrations of GSH and GSSG.…”

“…Mostly, spectrophotometric-and high-performance liquid chromatography (HPLC) based methods are used. Isotachophoresis (ITP) has proven to be a quick and cheap method for the detection of both GSH and GSSG (Bodor et al 2018). No standard method exists nowadays, therefore, comparing the results was difficult due to the different types of samples, the techniques used in the sample collection, preparation and measurements.…”

“…Electrochemical separation by isotachophoresis (ITP) seems to be another suitable method. It enables specific determination of both GSH and GSSG in the same sample at the same time (Bodor et al 2018). Nevertheless, no standard method of glutathione assessment exists and even discrepancies in sample preparation and units of GSH and GSSG levels aggravate the comparison of results among individual experiments.…”
