Carbohydrate status in dormant and afterripened excised wild oat embryos

Abstract: 1992. Carbohydrate status in dormant and afterripened excised wild oat embryos. -Physioi. Plant. 85; 461^66.Germination and carbohydrate concentrations were determined in excised dormant and afterripened wild oat (Avena fatua L. line M73) embryos cultured on N6 medium with and without 88 mM fructose (Fru). Without Fru dormant embryos began to germinate after approximately 2 weeks, and the germination rate was greater at 12 than 16'*C. With addition of Fru 80% of dormant embryos germinated in 3 days. More than … Show more

“…Sugar extraction followed the protocol of Foley et al (1992), except that ice‐cold 80% (v/v) ethanol and alternative internal standards were used. At the time of assay, samples were retrieved from −80°C storage and pulverized in liquid nitrogen in a mortar and pestle.…”

“…Soluble sugars are known to be able to overcome dormancy in seeds, e.g. in wild oat embryos (Foley et al 1992). The promoting effect of the sugars on germination may derive from the action of sugars as antagonists to other substances present in the tissues, which inhibit germination processes.…”

“…Shoot tip of these plants ca 3 leaves from the apex were then harvested 3 times, just after the application of fertilizer (T0), eleven day after the application (T1) and 24 days after the application (T2). Sugar in this plant materials were extracted using modified methods by Foley et al 1992 and sugar content in the mL extract were analyzed spectrophotometrically using Nelson-Somogie methods. This experiment was repeated using different plants with application of fertilizer in higher doses; 18.5, 2.2 and 9.2 g/plant for Urea, TSP and KCl, respectively.…”

“…These plants were sprayed with the fertilizers every 1-2 days for 132 days before its leaves were harvested for examination of sugar content. Sugar from this plants were extracted using modified methods by Foley et al 1992 and sugar content was analysed using Anthron methods.…”
