Carcinogen 7,12-dimethylbenz[a]anthracene-induced mammary tumorigenesis is accelerated in Smad3 heterozygous mice compared to Smad3 wild type mice

Abstract: Previous studies based on cell culture and xenograft animal models suggest that Smad3 has tumor suppressor function for breast cancer during early stages of tumorigenesis. In this report, we show that DMBA (7,12-dimethylbenz[a]anthracene), a chemical carcinogen, induces mammary tumor formation at a significantly higher frequency in the Smad3 heterozygous mice than in the Smad3 wild type mice. This is the first genetic evidence showing that Smad3 inhibits mammary tumor formation in a mouse model. Our findings s… Show more

“…In this context, 7,12‐dimethylbenzanthracene (DMBA) is a prokaryogen, which requires metabolic activation by cytochrome P450 (CVYP1A2 and CYP1A1, CYP1B1) enzymes to generate the metabolite dihydrodiolepoxide, which can generate DNA adducts in peripheral blood and breast tissue . It induces BC in animals by double strand breaks of DNA subsequent damage repair .…”

“…The induction of DMBA and Estradiol carcinogen compounds for 8 weeks induced the formation of breast cancer 7,12-Dimethylbenz[a]anthracene (DMBA) was a carcinogen compound that has an aromatic hydrocarbon chain and has been widely used in research as an inducer in breast cancer modeling [22,23,24]. Estradiol is a steroid the sex hormone that useful in female reproductive cycle in regulation of estrous and menstrual cycles.…”
