Card9- and MyD88-Mediated Gamma Interferon and Nitric Oxide Production Is Essential for Resistance to Subcutaneous Coccidioides posadasii Infection

Abstract: Coccidioidomycosis is a potentially life-threatening respiratory disease which is endemic to the southwestern United States and arid regions of Central and South America. It is responsible for approximately 150,000 infections annually in the United States alone. Almost every human organ has been reported to harbor parasitic cells of Coccidioides spp. in collective cases of the disseminated form of this mycosis. Current understanding of the mechanisms of protective immunity against lung infection has been large… Show more

“…Several studies support the concept that CARD9 contributes to the innate immune system's response to infectious diseases through the recognition of bacterial, viral and fungal pathogens [12][13][14]. A deficiency of CARD9 failed to trigger an innate host defence reaction against microbial infections.…”

“…46 Murine data suggest that IL-4 itself directly reduces the antifungal activity of innate immune cells as it promotes expression of arginase-1 and blocks nitric oxide production, 47 which is required for fungicidal activity. 48 Murine studies of cryptococcal infection indicate that IL-13 works in a similar fashion 49 in promoting high expression of IL-4.…”

“…In contrast, NOS2 is widely distributed in multiple cell types, and is significantly induced under certain infection or inflammatory stimulations via PAMPs [17,18]. For example, microbe-induced NOS2 production can be facilitated by myeloid differentiation factor 88 and the caspase adaptor recruitment domain family member-9-mediated nuclear factor (NF)-κB signaling pathway in a calcium-independent manner [19,20].…”
