Cardiac Valve Bioreactor for Physiological Conditioning and Hydrodynamic Performance Assessment

Abstract: Purpose: Tissue engineered heart valves (TEHV) are being investigated to address the limitations of currently available valve prostheses. In order to advance a wide variety of TEHV approaches, the goal of this study was to develop a cardiac valve bioreactor system capable of conditioning living valves with a range of hydrodynamic conditions as well as capable of assessing hydrodynamic performance to ISO 5840 standards. Methods: A bioreactor system was designed based on the Windkessel approach. Novel features i… Show more

“…All three parameters are important indicators for the clinical assessment of aortic valve stenosis severity 28,[59][60][61] . Although the baseline hydrodynamic performances of unimplanted BHV in pulse simulators have been repeatedly reported [62][63][64] , to the best of our knowledge, we are the first to evaluate the hydrodynamic performances of BHV following in vitro incubations that simulate AGE formation observed in clinical explants. In general, all in vitro incubation conditions resulted in a steady decline in EOA and increases in pressure gradients and peak jet velocities.…”

“…A schematic of the flow chamber is shown in Figure 4. Test chambers with similar geometry are seen in valve testing devices [56] and standards [23], and have been shown to be sufficient for valve testing in in vitro experiments [45,44,55,37,11]. The domain is divided into 96 × 96 × 192 points for a spatial resolution ∆x = 0.046875 cm in the fluid.…”

“…None of the studies assessed in this article combined a recellularization component with decellularization in their tissue-engineering technology. Much work is being undertaken to explore the capabilities of tissue-engineered scaffolds that are produced from decellularized xenografts and then recellularized using the future host's autologous cells [23,24,32]. Such a graft is expected to be living and thus have the potential to grow, repair, and remodel in situ [15][16][17][18][19].…”

“…In addition, a recent concept being explored in heart valve tissue-engineering is construct recellularization using autologous cells [ 32 ]. In essence, a decellularized construct is repopulated with the recipient’s cells and then conditioned within a bioreactor ahead of surgical implantation [ 23 , 24 ]. None of the studies assessed in this article combined a recellularization component with decellularization in their tissue-engineering technology.…”
