Cardioactive C19-Diterpenoid Alkaloids from the Lateral Roots of <i>Aconitum carmichaeli</i> “Fu Zi”

Abstract: Bioassay-guided fractionation of an n-BuOH extract of the lateral roots of Aconitum carmichaeli DEBX. led to the isolation of 5 cardioactive C 19 -diterpenoid alkaloids: N-deethylaconine (1), beiwutinine (2), hypaconine (3), mesaconine (4), and 15α-hydroxyneoline (5). N-Deethylaconine and beiwutinine are new aconitinetype C 19 -diterpenoid alkaloids. Hypaconine was isolated from this species for the first time. Among them, mesaconine, hypaconine, and beiwutinine showed the strongest cardiac actions on the isol… Show more

“…For hypaconitine, the retention time was found to be 14.4 min with a m/z ratio of 616.311. The other component peaks were identified based upon the m/z values and information available in literature [27,35-37]. To exclude the interferences of the endogenous and exogenous substances that can be present in filtrate of cow urine and cow milk after extraction, the list of components identified in the blank controls with their m/z values, retention times, and masses are mentioned in Additional file 5: Table S2 (additional data file).…”

“…In the MS 3 spectrum, two expected characteristic ions at m/z 508 and 498, derived from neutral loss of 50 Da (MeOH+H 2 O) and 60 Da (MeOH+CO) respectively, were observed. Based on its fragmentation pattern (Figure 8), Peak 12 was preliminarily identified as N-deethyl-aconitine, the de-esterification product of which has been reported from A. carmichaeli [36].…”

“…Modern pharmacological researches show that medicinal plants in Genus Aconitum have an extensive pharmacological action and clinical application such as anti-inflammation,[1] immunosuppression,[2] narcotic analgesic action,[3] anti-tumor, and so on. [4] Due to the medical significances of plants in Aconitum L., in-depth exploration and research around contend determination of alkaloids, chemical structure analysis and determination of bioactivity have been conducting for a long time.…”
