Cardiovascular Metabolomics

Abstract: Disturbances in cardiac metabolism underlie most cardiovascular diseases. Metabolomics, one of the newer omics technologies, has emerged as a powerful tool for defining changes in both global and cardiac-specific metabolism that occur across a spectrum of cardiovascular disease states. Findings from metabolomics studies have contributed to better understanding of the metabolic changes that occur in heart failure and ischemic heart disease and have identified new cardiovascular disease biomarkers. As technologi… Show more

“…(21,22) More importantly, cardiac metabolism is altered in the presence of heart failure due to decreased mitochondrial function as a result of the accumulation of medium and long chain acyl-carnitine, increased ketone metabolism and the accumulation of lactate and branch chain amino acids. (23) Patients with diabetes, therefore, are at increased risk for the development of heart failure and the prevention of heart failure is becoming a major emerging research and treatment goal such as the discovery of the sodium-glucose co-transporter 2 (SGLT2) inhibitors for the treatment of diabetes and coexistent heart failure. (24) In addition to the admission value, a graded relationship has been found between increasing blood glucose levels and MACE.…”

“…Several metabolic dysregulations occurring in cardiomyocytes as well as in other tissues were associated with a large spectrum of CV diseases. 85 The human metabolome refers to the complete set of small molecules including endogenous metabolites (eg, amino acids, organic acids, nucleic acids, fatty acids, amines, sugars, vitamins, co-factors, pigments, antibiotics, etc) and exogenous chemicals (such as drugs, environmental contaminants, food additives, toxins and other xenobiotics) as products of metabolic reactions on July 7, 2020 by guest. “…86 All of these metabolites can be measured by using different advanced metabolomics platforms including nuclear magnetic resonance (NMR spectroscopy), mass spectrometry (MS), liquid chromatography (LC), gas chromatography or capillary electrophoresis to facilitate compound separation. 85 Basic findings from metabolomics applications are contributing to better clarifying the metabolic unbalance that occurs in CV diseases suggesting useful non-invasive circulating biomarkers. Moreover, Würtz et al 87 conducted NMR metabolomics to three large cohorts in order to identify circulating biomarkers of incident CV events.…”

“…Molecular mechanism investigation provides useful information related to diseases that could be used in their management [3]. High throughput methods such as proteomics, genomics, and metabolomics are the advanced method used widely to investigate molecular aspects of many diseases [4][5][6]. In the metabolomics studies, differentially levels of metabolites such as lipids, amino acids, and organic acids in the patient samples rel-GMJ.2020;9:e1696 www.gmj.ir ative to the controls are evaluated [7].…”
