Carrageenan‐induced inflammation promotes ROS generation and neutrophil extracellular trap formation in a mouse model of peritonitis

Abstract: Neutrophil extracellular traps (NETs) are a combination of DNA fibers and granular proteins, such as neutrophil elastase (NE). NETs are released in the extracellular space in response to different stimuli. Carrageenan is a sulfated polysaccharide extracted from Chondrus crispus, a marine algae, used for decades in research for its potential to induce inflammation in different animal models. In this study, we show for the first time that carrageenan injection can induce NET release in a mouse model of acute per… Show more

“…The doses of sterile saline, BCP, and DHA mentioned above were administered subcutaneously 30 min before intraperitoneal injection of C g (500 μg / animal) into their respective groups. After 4 h, the mice were euthanized and their peritoneal cavity was washed with 3 mL of phosphate‐buffered saline (PBS) containing 1 mM ethylenediaminetetraacetic acid (EDTA) . From the volume removed, an aliquot of 0.02 mL was added to 0.4 mL of Turk's solution for counting the total number of leukocytes in a Neubauer chamber (PRESVAC® CT12, Curitiba, Brazil).…”

The differential count (100 cells) was performed on a cytocentrifuge, prepared and stained with Diff-Quik.…”

“…18 Further, carrageenan-evoked inflammatory action is associated with increased NOX2/ROS production in phagocytes. 19,20 Thus we tested VIP effect on carrageenan-induced acute paw edema in rats. The 5-week-old rats that received carrageenan in their right paws had a progressive increase in the thickness of edema as compared with their saline-injected left paws (Figure 6a).…”

“…Recent studies have revealed that neutrophil extracellular traps (NET), which consist mainly of the DNA-histone complex and neutrophil elastase, can directly activate factor XII (5,6). Considering that thyroid hormone induces reactive oxygen species (ROS) production in neutrophils and ROS can induce NET formation (7,8), it is plausible that NET may be actively formed in hyperthyroidism. NET can activate the contact system (3,5).…”
