Cathepsin V, a Novel and Potent Elastolytic Activity Expressed in Activated Macrophages

Abstract: Atherosclerosis is characterized by a thickening and loss of elasticity of the arterial wall. Loss of elasticity has been attributed to the degradation of the arterial elastin matrix. Cathepsins K and S are papain-like cysteine proteases with known elastolytic activities, and both enzymes have been identified in macrophages present in plaque areas of diseased blood vessels. Here we demonstrate that macrophages express a third elastolytic cysteine protease, cathepsin V, which exhibits the most potent elastase a… Show more

“…The findings of this study indicate the novel role of cathepsin V in secretory vesicles for producing active neuropeptides, which contrasts with the well known function of cathepsin V in lysosomes (11,45). This study shows that human neuroblastoma cells contain cathepsin V in secretory vesicles as well as lysosomes.…”

“…After protein transfer and blocking of nonspecific binding sites for 2 hours with 5% skimmed milk in PBST, membranes were incubated overnight at 4°C with the rabbit polyclonal antibodies to cath-D (1: 7500), 22 cath-B (both 1:500), and cath-H (1:10,000) and mAbs to cath-K (1:200), cath-V (1:500). Also, we used the two mAbs specific for procath-L (2D4 at 1:500) and all three cath-L forms (33/1 at 1:500), 23,24 respectively. Secondary horseradish peroxidase-conjugated goat antirabbit and rabbit anti-mouse Igs were used at 1:20,000 for 1 hour at room temperature, respectively.…”

“…MMPs degrade fibrillar elements such as fibrillar collagens and elastin, and nonfibrillar elements (nonfibrillar collagens, glycoproteins, and proteoglycans) such as type IV collagen, the proteoglycan perlecan or glycoproteins laminin and entactin, all constitu- 43 Recently, cathepsin V has been identified in atherosclerotic plaque specimens and exhibits the most potent elastase activity yet described among human proteases. 35 The plasminogen system degrades laminin, fibronectin, vitronectin, and proteoglycans. Neutrophil elastase degrades collagen, elastin, laminin, fibronectin, and proteoglycan.…”
