CCR7/CCL21 Migration on Fibronectin Is Mediated by Phospholipase Cγ1 and ERK1/2 in Primary T Lymphocytes

Abstract: CCR7 binds to its cognate ligand, CCL21, to mediate the migration of circulating naive T lymphocytes to the lymph nodes. T lymphocytes can bind to fibronectin, a constituent of lymph nodes, via their ␤1 integrins, which is a primary mechanism of T lymphocyte migration; however, the signaling pathways involved are unclear. We report that rapid (within 2 min) and transient phosphorylation of ERK1/2 is required for T cell migration on fibronectin in response to CCL21. Conversely, prevention of ERK1/2 phosphorylat… Show more

“…CCL19 and CCL21: ligands of CCR7; CCL20: ligand of CCR6 CCL19/CCR7 has been implicated in the immune surveillance of the CNS by lymphocytes Shannon et al 2010). CCL21 is rapidly upregulated in cortical neurons upon ischaemic injury and glutamate-mediated excitotoxicity, is transported in secretory granules along axons to presynaptic structures and, in this way, activates remote microglia expressing CXCR3 (Biber et al 2001(Biber et al , 2011 for the role of CXCR3 in CNS inflammatory reactions, see above).…”

“…Stable expression of a PLCγ1 PH domain fragment blocks migration [162]. PLCγ1 mediates CCL21-induced T cell migration on fibronectin through cross-talk with ERK1/2 [163]. It also promotes Rac activation, invasion and metastasis in breast cancer cells [164].…”

“…Furthermore, ERK1/2 is required for the migration of naïve T lymphocytes toward CCL21, another ligand of CCR7. 32 We hypothesized that ZAP70 activates MAPK pathways to regulate CCR7 and CXCR4 and to mediate CCL19-and CXCL12-induced migration. In order to examine ZAP70 expression and ERK activation, we analyzed 9 patient xenografts by western blotting ( Figure 3A, left).…”
