CD209 (DC-SIGN) −336A>G promoter polymorphism and severe acute respiratory syndrome in Hong Kong Chinese

Abstract: CD209 (DC-SIGN) is an important C-type lectin which acts a receptor of many pathogens. The single nucleotide polymorphism (SNP) -336A>G in the CD209 promoter has been demonstrated to regulate promoter activity and to be associated with several important infectious diseases, such as human immunodeficiency virus-1 (HIV-1), Mycobacterium tuberculosis, and Dengue fever. CD209 facilitates severe acute respiratory syndrome (SARS)-coronavirus spike protein-bearing pseudotype driven infection of permissive cells in vi… Show more

“…Based on this hypothesis, we examined the influence of five single nucleotide polymorphisms (SNPs) in the promoter region of DC-SIGN that have been reported to a) influence the promoter activity and b) are associated with several infectious diseases [13,14,[17][18][19][20][21][22], with IVIG response among KD patients of three ethnic groups -Whites, Asians and Hispanics. KD patients who were treated with IVIG and had documented response data from our ongoing cohort recruited from Seattle Children's Hospital, Oakland Children's Hospital, and Primary Children's Hospital of Utah were included in this study.…”

“…For example, SNPs of genes involved in innate immunity were correlated with SARS-CoV load during the initial phase of infection [49]. A 336A > G promoter polymorphism in the gene encoding CD209 (DC-SIGN) was correlated with clinical-pathologic outcomes in 824 serologically confirmed SARS patients [50]. The -336AG/GG genotype was associated with lower standardized lactatedehydrogenase (LDH) levels compared with the '-336AA' genotype carrying patients with a 60% chance of having a poorer prognosis because of higher LDH levels.…”

“…Moreover, M tuberculosis Manose-dependent binding to CD209 leads to Raf-1-dependent upregulation of IL-6 [15]. However, the DC-SIGN -336G variant affects a Sp1 binding site and decreases the expression of DC-SIGN resulting in less efficient cytokine and chemokine secretion [8,30]. It is hypothesized that individuals homozygous for the -336G allele may, in part, be protected from expressing low levels of DC-SIGN and therefore less prone to the IL-6 secretion induced by Manose-dependent binding to DC-SIGN.…”
