CD47 but Not Calreticulin Is over Expressed in Patients with Myeloproliferative Neoplasms

Abstract: Myeloproliferative neoplasms (MPN) are chronic myeloid cancers characterized by the overproduction of mature blood cells, and the tendency to evolve into acute leukaemia. In solid tumours, calreticulin (CALR) overexpression on the cell surface, produces a pro-phagocytic signal and is counteracted by concomitant expression of anti- phagocytic CD47, reflecting an apoptosis vs survival mechanism. In this study, we investigated the expression of CALR and CD47 in patients with MPN and potential changes induced by t… Show more

“…The use of targeting antibodies against CD47 are currently under development for the treatment of AML and MDS, 81,82 with demonstrated efficacy in combination with azacitidine including in patients with TP53‐mutant AML who are generally refractory to standard therapies. CD47 is also overexpressed in patients with MPNs and TP53 is a common driver of leukaemic transformation in patients with MPNs, suggesting potential utility here too 83,84 …”

“…Another strategy that led to further improvements in anti-47 therapy was through combination with other molecules, which helped unmask or increase prophagocytic signals in leukemia cells in the setting of removal of the "do not eat me" signal. Calreticulin is one such pro-phagocytic signal and its expression is upregulated by exposure to azacitidine in MDS and MPN cell lines [22]. Clinical benefit has been demonstrated in patients with TP53-mutated AML.…”
