Cd8 T‐cell recognition of human 5T4 oncofetal antigen

Abstract: The 5T4 oncofetal antigen is expressed by a wide variety of human carcinomas, including colorectal, ovarian and gastric carcinomas. The restricted expression of 5T4 on tumor tissues as well as its implication in tumor progression and bad prognosis makes 5T4 a promising new candidate for immunotherapy. An MVA vaccine encoding 5T4 antigen has been successfully evaluated in preclinical studies in a murine tumor model. Here, we report the generation of human CD8 T cells specific for the 5T4 antigen by stimulation … Show more

“…5T4-specific CD8 + T-cells have been generated against 5T4 sequences before [25]. High affinity T-cell receptor binding of the 17-25 amino-acid sequence of 5T4 has been demonstrated (ED50 1.5 × 10 −7 M), and strong ELISPOT responses were elicited against this peptide in a patient vaccinated with TroVax ® [19].…”

“…Antigens and cellular responses. To explore changes in in vitro responses to relevant TAAs, overlapping 5T4 32-mer peptides (18 patients, 24 cycles) and purified CEA (13 patients, 15 cycles) were used in a thymidine incorporation proliferation assay (26). All time points for each patient were investigated in the same assay: cryopreserved PBMCs were thawed and 1.5 × 10 5 viable cells per well were cultured in Iscove's modified Dulbecco's medium with 10% AB serum, with six pools of four overlapping 5T4 peptides (defined as A-X) spanning the whole sequence of human 5T4 (10 μg/mL of each peptide); media controls with and without the appropriate concentration of dimethyl sulfoxide were used to prepare peptide stocks (27,28).…”
