cDNA clone for the human invariant gamma chain of class II histocompatibility antigens and its implications for the protein structure.

Abstract: The invariant y chain is transitorily associated with class H histocompatibility antigens during intracellular transport. We have isolated and sequenced a DNA clone corresponding to the human y chain. mRNA hybridizing to the cDNA clone translated into a 33,000-dalton chain that associated specifically with class H antigen a and f3 chains. The y chain consists of 216 amino acids. The two N-linked carbohydrates are attached to asparagines 114 and 120. A continuous stretch of hydrophobic and neutral amino acids o… Show more

“…From the DNA sequence, p35 differs from p33 in having an additional 16-amino acid NH2-terminal segment. Very little is known concerning the human p41 protein, except that it is also observed in cell-free translation products of In mRNA purified by hybridization selection (Claesson et al, 1983;and unpublished). In mouse, it has been shown that p41 is structurally related to In p33 (Zecher et al, 1984).…”

“…We tested the effects of CD23-mediated B cell activation on surface expression of costimulatory molecules, CD86 and CD80, as well as HLA-DR but found no effect (not shown). The HLA-DR-associated invariant chain, CD74, is a MHC class II chaperone and plays a role in Ag presentation (24). CD74 and HLA-DR are associated at the cell surface.…”

“…CD74 (invariant chain) is a type-II transmembrane protein, with 30 NH 2 -terminal, intracytoplasmic amino acid residues, a membrane-spanning segment from residues 31 to 55, and an extracytoplasmic domain consisting of 160 amino acids (1,2). Once synthesized, CD74, DR␣, and DR␤ begin to associate within the endoplasmic reticulum.…”
