cDNA sequence of rat liver fructose-1,6-bisphosphatase and evidence for down-regulation of its mRNA by insulin.

Abstract: A coding-length clone of rat liver fructose-1,6-bisphosphatase (EC 3.1.3.11) was isolated by immunological screening of a cDNA library in Agtll. Its identity was verified by comparing the deduced amino acid sequence with that obtained by direct sequencing of a complete set of CNBr and proteolytic peptides from the purified protein. The enzyme subunit is composed of 362 amino acids and has N-acetylvaline as the amino-terminal residue. The cDNA, 1255 base pairs (bp) long, consisted of 1086 bp of coding region, 1… Show more

“…Secondly, all C/EBP-family members do not display the same transcriptional properties, and most often the C/EBP isoform was not identified. Previous reviews [3,4] mouse liver [155,156] and decreases FBPase-l mRNA in rat liver [157,158] (Figure 4). We (Table 3).…”

“…The sequence data obtained from the 40-kDa protein corresponded to the sequence of a mixture of two peptides derived from rat liver AI, 28) while the data from the 42-kDa protein corresponded to the sequence of a mixture of three peptides derived from rat liver FBPase. 29) All the detected peptide sequences began from the proline residue following the aspartic acid residue in their original proteins, indicating that these peptides were produced by specific cleavage at the acid-labile Asp-Pro bonds. These results indicate that the 40-kDa and 42-kDa proteins were AI and FBPase respectively.…”

“…Each identical subunit has 337 amino acid residues and a molecular weight of about 36,500 (Marcus et al, 1982;Williams & Kantrowitz, 1992). Mammalian enzymes (liver and kidney) are more than 85% homologous in amino acid sequence (El-Maghrabi et al, 1988). The physiologic regulators AMP and Fru-2,6-P2 negatively modulate enzyme activity and, in reciprocal fashion, positively affect the activity of phosphofructokinase, a control point in glycolysis (for a review, see Pilkis & Claus, 1991).…”
