Celestrol-Loaded Nanoscale Micelles Derived from Dextran Sulfate/PVGLIG/Celestrol with MMP-2 Sensitivity for the Treatment of High-Fat-Diet-Induced Obesity

Abstract: With the improvement of people’s living standards, obesity has become a health threat, but an effective drug treatment for obesity has not been found yet. Here, we suggest the use of celastrol, a pentacyclic triterpenoid from the traditional Chinese medicine Tripterygium wilfordii, which has a powerful antiobesity effect. Increasing studies have discovered that obesity is closely related to inflammation. In addition, MMP-2 enzyme is highly expressed in inflammatory sites. According to the principle of the prod… Show more

“…The fat liver comorbidity symptoms were significantly alleviated, and little weight regain was associated after treatments. Compared with the previously reported obesity treatment efficacies that were achieved via regulation of signal pathway for fat metabolism, immune regulation, reprogramming of intestine microbiota, browning white adipocytes or photothermal ablation, the weight loss achieved via current strategy of camouflaging adipocytes with apoptotic feature is remarkable (33.3%) and comparable with the efficacy achieved by the well‐known counterpart, such as pro‐apoptosis peptide KLA (≈30%), [ 19 ] celastrol (≈54%), [ 54 ] rosiglitazone (≈15%), [ 13 ] capsaicin (22.4%), [ 55 ] photothermal agents (≈15% for Copper sulfide hydrogel), [ 56 ] and etc. (Table S1, Supporting Information).…”
