Cell Biology of the Glomerular Podocyte

Abstract: Glomerular podocytes are highly specialized cells with a complex cytoarchitecture. Their most prominent features are interdigitated foot processes with filtration slits in between. These are bridged by the slit diaphragm, which plays a major role in establishing the selective permeability of the glomerular filtration barrier. Injury to podocytes leads to proteinuria, a hallmark of most glomerular diseases. New technical approaches have led to a considerable increase in our understanding of podocyte biology inc… Show more

“…Herein, we also demonstrated that 1,25(OH)2D3 treatment in nephropathy rats could up-regulate the expressions of nephrin and podocin, α3β1 integrin and α/β-DG, and decrease podocyte loss and FPW, which may lead to improvement in podocytopenia, proteinuria and a decrease in the urinary podocyte number. It has been shown that proteinuria is associated with foot process effacement (25). Moreover, podocyturia is a more sensitive indicator in assessing the degree of glomerular damage than proteinuria in SD rats (26).…”

“…This filtration takes place in the glomerulus, a specialized knot of blood vessels whose wall forms the filtration barrier and is composed of three layers: the endothelium, glomerular basement membrane, and visceral epithelial cells or podocytes. Podocytes have finger-like projections, called pedicels, or foot processes, that interdigitate with those from neighboring cells and that are separated by a glycoprotein network forming slit pores of~40 nM in width (1). Although the role of these slit pores in filtration is conjectural, there is clear evidence that changes in podocyte morphology are associated with changes in the integrity of the glomerular filter; for example, the loss of podocytes and progressive nephropathy of diabetes (2).…”

“…Although the role of these slit pores in filtration is conjectural, there is clear evidence that changes in podocyte morphology are associated with changes in the integrity of the glomerular filter; for example, the loss of podocytes and progressive nephropathy of diabetes (2). Importantly, congenital forms of nephrotic syndrome are associated with mutations of podocyte-specific genes (1). The limited ability of the podocyte to repair and/or regenerate is believed to be crucial in progressive renal disease (3).…”

“…Podocytes are localized in the outer aspect of the basement membrane, and injury of these cells is known to be associated with proteinuria. 16 Cultured podocytes exposed to mechanical stress enhance angiotensin II type 1 receptor gene expression, which results in podocyte injury. 17 In the present study, the desmin staining score, which shows the degree of damaged podocytes, was significantly reduced by irbesartan.…”
