Cell cycle– and swelling-induced activation of a Caenorhabditis elegans ClC channel is mediated by CeGLC-7α/β phosphatases

Abstract: ClC voltage-gated anion channels have been identified in bacteria, yeast, plants, and animals. The biophysical and structural properties of ClCs have been studied extensively, but relatively little is known about their precise physiological functions. Furthermore, virtually nothing is known about the signaling pathways and molecular mechanisms that regulate channel activity. The nematode Caenorhabditis elegans provides significant experimental advantages for characterizing ion channel function and regulation. … Show more

“…In oocytes, the activity of CLH-3b is strongly inhibited by phosphorylation by the GCK-3 kinase (Denton et al, 2005) and activated by the GLC-7␣/␤ serine/threonine phosphatases (Rutledge et al, 2002). The activity of CLC-2 is also regulated by phosphorylation (Rutledge et al, 2002) and by PKC (Madison et al, 1986). In the future, identifying the signaling pathways upstream of clh-3 in the HSN might provide a window into the contexts in which this family of channels acts to modulate neural activity.…”

“…Like CLC-2, the clh-3-encoded channel variants CLH-3a and CLH-3b are swelling-and hyperpolarization-activated inwardly rectifying chloride channels. CLH-3b is expressed in C. elegans oocytes, where it is activated during meiotic maturation and plays a role in regulating ovulation (Rutledge et al, 2001). The in vivo role of the CLH-3a variant is not known.…”

“…ClC-2 interacts with PP1 isoforms in the yeast two-hybrid system, and ClC-2 currents that were increased by the cyclindependent kinase inhibitor olomoucine were again reduced by the PP1/PP2A inhibitor calyculin A (140). Moreover, it has recently been shown that the maturation and swelling-induced activation of the Caenorhabditis ortholog of ClC-2 during meiosis is mediated by PP1 (308). Interestingly, the phenotype of ClC-2 knock-out mice (56) is reminiscent of that of PP1␥ null mice (376); both mice combine a normal gross anatomy with exclusively male infertility, which is correlated with a reduced diameter of the seminiferous tubules and a loss of germ cells.…”

“…To determine whether CLH-3b⌬C channels are functional, we recorded whole cell Cl Ϫ currents in oocytes isolated from wild-type and clh-3(ok768) worms. As shown previously (9,27,28), wild-type meiotic cell cyclearrested, nonswollen oocytes exhibit a small inwardly rectifying CLH-3b current (I CLH-3b ) that is activated by strong hyperpolarization. Figure 2A shows mean whole cell current traces recorded in nine wild-type oocytes.…”

“…clh-3 RNAi disrupts CLH-3 channel expression in the C. elegans oocyte (9,27,28). Steady-state current amplitude recorded at Ϫ100 mV in oocytes isolated from clh-3(ok768) worms injected with clh-3 dsRNA was inhibited Ͼ95% (P Ͻ 0.01) to a mean Ϯ SE value of Ϫ1.2 Ϯ 0.8 pA/pF (n ϭ 3).…”
