Cell surface and relative mRNA expression of heat shock protein 70 in human synovial cells

Abstract: Hsp70 gene expression in RA-affected synovial tissue is followed by Hsp70 cell surface expression on fibroblast-like synovial cells growing from RA synovial tissue. Hsp70 may be translocated to the cell surface from the cytosol and/or Hsp70 released from inflamed synovial tissue may be captured onto the membrane of synovial cells from the extracellular space via Hsp receptors. As a physiological response to potentially harmful enviromental stress factors, skin dermis produces higher levels of Hsp70 comparing t… Show more

“…Health factors possibly associated with HSP70 were assessed via face-to-face interview and data abstraction from medical chart reviews. Health factors assessed included length of time since abnormal endometrial biopsy was obtained; body mass index (BMI) (Ghayour-Mobarhan et al, 2007); tumor cellular classification, grade, and stage (Nanbu et al, 1998); and comorbid connective tissue disease (Sedlackova et al, 2009). Medical comorbidity severity was assessed using the Charlson Comorbidity Index (Charlson et al, 1987).…”

“…The observation of elevated levels of hsp70 in synovial fluids from inflamed joints of RA patients is one of these factors. Hsp70 is found both within the fibroblasts at the joint and in the fluid itself [99, 100]. The significant increase in extracellular hsp70 in arthritic joints is profoundly correlative because nonarthritic joints in the same individual patients have no elevation in hsp70.…”

“…Furthermore, in patients with RA or JIA, augmented Hsp expression has been observed in the inflamed synovium and found to be important for proper functioning of Hsp-specific regulatory T cells (3). Our group previously reported higher expression of Hsp70 in synovial cells derived from patients with severe RA and JIA (4).…”
