Cell therapy to improve regeneration of skeletal muscle injuries

Abstract: Diseases that jeopardize the musculoskeletal system and cause chronic impairment are prevalent throughout the Western world. In Germany alone, ~1.8 million patients suffer from these diseases annually, and medical expenses have been reported to reach 34.2bn Euros. Although musculoskeletal disorders are seldom fatal, they compromise quality of life and diminish functional capacity. For example, musculoskeletal disorders incur an annual loss of over 0.8 million workforce years to the German economy. Among these … Show more

“…Moreover, muscular traumas, often during sports performance, are very common evidence associated with high medical expenses and disabilities. In addition to promising clinical trials with progenitor cells in humans [105], there are convincing data on the beneficial role of melatonin in crushed injured muscles in rodents. Chronic melatonin intake reduced apoptosis, increased twitch force, and accelerated regeneration enhancing satellite cells in muscle injury in mice and in rat [106,107].…”

“…Their mode of action has mainly been considered to be due to secretion of a variety of cytokines and growth factors that can promote angiogenesis, cell recruitment, migration, proliferation, and differentiation [5]. The general immunomodulatory capacity of stromal cells has also been hypothesized to exert beneficial effects on the local and/or systemic immune response [5], [6].…”

“…In this study we focused on extracellular vesicles (EVs) compared to secreted soluble factors as potential mediators of PLX's biologic activity. 5 October 17,2019 In initial experiments, we compared the particle count of standard media used at our institution for stromal cell culture to select a suitable medium for PLX cell-derived EV isolation. Alpha-modified minimum essential medium (-MEM) as well as other conventional media supplemented with either fetal bovine serum (FBS) or pooled human platelet lysate (HPL) as published [13] contained mean 4 x 10 8 -3 x 10 9 particles/mL.…”

“…Many studies to date have explored the ability of MSCs to support bone development, restoration of ventricular functional, and improved renal tubular function in vivo and in clinical settings. [4][5][6] Unfortunately, however, these cells are limited in their therapeutic efficacy, particularly in contexts where injuries or the associated ischemic damage are severe and irreversible. Indeed, preclinical animal models suggest that MSCs have a poor ability to engraft, and they are also hampered by limited homing and survival in vivo owing to factors including inflammation, ischemia, and anoikis.…”
