Cellular and subcellular localization of Huntington aggregates in the brain of a rat transgenic for Huntington disease

Abstract: Huntington disease (HD) is a progressive neurodegenerative disorder characterized by emotional, cognitive, and motor dysfunctions. Aggregation of huntingtin is a hallmark of HD and, therefore, a crucial parameter for the evaluation of HD animal models. We investigated here the regional, cellular, and subcellular distribution of N-terminal huntingtin aggregates and associated neuropathological changes in the forebrain of a rat transgenic for HD (tgHD). The tgHD rat brain showed enormously enlarged lateral ventr… Show more

“…Dark degenerating neurons, or dark neurons, have been detected in various pathological conditions including ischemia, epilepsy, 73 and chronic neurodegenerative states, such as Huntington's disease and models of this disease. 74,82,83 Our data provide clear evidence that caspase-3 is minimally activated in neurons undergoing dark cell degeneration; however, it remains to be investigated whether or not dark cell degeneration leads to neuron loss and whether it represents a distinct type of neuronal cell death, a stage of apoptosis preceding the activation of caspase-3, or a form of apoptosis involving a different set of caspases. The existence of at least two modes of neurodegeneration in the same mouse model underscores the complexity of cell death patterns in chronic neurodegenerative disease.…”

“…Electron microscopic sections were analyzed to compare the cellular integrity of PV neurons showing either low or high expression of the PV protein and to detect neuronal damage. A detailed description of the method has been published previously (Petrasch-Parwez et al, 2004, 2007. Sections for immunohistochemistry were reduced in 1% NaBH 4 in PBS for 30 min and incubated freely floating for 30 min in PBS containing 10% normal NGS.…”

“…and transcardially perfused by a 4% PFA. Fixed brains were removed from the skull, coronally cut into 50 m vibratome sections, and immunostained as recently described (Petrasch-Parwez et al, 2007). Briefly, after reduction and blocking, sections were incubated for 72 h at 4°C with anticalbindin D-28k (1:2000) (CB-38a; Swant, Bellinzona, Switzerland), anti-tyrosine hydroxylase (TH) (1:1000) (AB 152; Chemicon, Hofheim, Germany), or 15G7-␣-syn (1:10) (Kahle et al, 2000) in 10% normal goat serum or 10% NRS.…”
