Cellular localization and functional expression of P‐glycoprotein in rat astrocyte cultures

Abstract: We investigated the cellular/subcellular localization and functional expression of P-glycoprotein, an ATP-dependent membrane-associated efflux transporter, in astrocytes, a brain parenchyma compartment that is poorly characterized for the expression of membrane drug transporters. Analyses were carried out on primary cultures of astrocytes isolated from the cerebral cortex of neonatal Wistar rats and CTX TNA2, an immortalized rat astrocyte cell line. Both cell cultures display morphological features typical of … Show more

“…However, different cell types used for these experiments may account for the different results. Although the majority of data in literature indicates the localization of P-gp in caveolae (Rolfe et al, 2005;Ronaldson et al, 2004), other reports suggest lipid rafts but non-caveolar localization of this efflux transporter (Radeva et al, 2005) or localization both in caveolae and non-caveolae membrane domains (Jodoin et al, 2003). Lipid rafts are membrane microdomains that are enriched with cholesterol; thus, cholesterol depletion by MbCD is an effective strategy to disrupt lipid rafts.…”

“…Our laboratory has also characterized P-gp functional expression in a rat microglia cell line (MLS-9) (Lee et al, 2001), cultured rat astrocytes (Ronaldson et al, 2004a), and in an immortalized rat brain microvessel endothelial cell line (RBE4) (Bendayan et al, 2002).…”
