Change of drug resistance patterns and genetic properties of R plasmids inSalmonella typhimuriumof bovine origin isolated from 1970 to 1979 in northern Japan

Abstract: SUMMARYA total of 321 Salmonella typhimurium strains of bovine origin obtained in northern Japan during the period [1970][1971][1972][1973][1974][1975][1976][1977][1978][1979] were tested for drug resistance and detection of conjugative R plasmids. Three hundred and eighteen (99-1 %) of these strains were resistant to one or more drugs. The isolation frequency of multiply drug-resistant strains tended to increase year by year. Two hundred and thirtyseven (74'5 %) of these resistant strains carried conjugative … Show more

“…This study shows that the incidence of plasmids, drug resistance, and conjugative R plasmids was extraordinarily high in S. typhimurium strains isolated in Japan. Similar data on Salmonella isolates including S. typhimurium have been published elsewhere (8,22). We isolated S. typhimurium strains from apparently healthy male calves and found a high incidence of multiple drug resistance and plasmids of various sizes (10).…”

“…To clarify whether or not S. Typhimurium isolates have the ability to transfer their resistance genes, liquid and filter mating procedures were performed in vitro (21) using rifampicin-resistant Escherichia coli C600 (Rif r ) and Nal-resistant S. Dublin (Nal r ) derivatives as recipient strains. S. Typhimurium isolate No.7 was used as the donor strain.…”

“…Large plasmid DNA was isolated and agarose gel electrophoresis was performed as previously described [4]. The R plasmids were detected by filter mating at 37 mC using E. coli C600 Rif r as the recipient strain and Tet r O157 isolates as the donor strains as previously described [5]. DNA sequencing was performed using an ABI PRISM 377 (Applied Biosystems).…”
