During surveys conducted in 2012–2013, viruslike symptoms of chlorotic spots with, in some cases, a necrotic centre in older leaves were observed in field‐ and greenhouse‐grown cucumber (Cucumis sativus L.), melon (C. melo L.) and squash (Cucurbita sp.) in the major cucurbit cultivation regions in Iran. Leaf samples were collected and tested for the presence of Cucumber leaf spot virus (CLSV, genus Aureusvirus, family Tombusviridae) by a virus specific double‐antibody sandwich enzyme‐linked immunosorbent assay (DAS‐ELISA). CLSV was detected in four of eight surveyed provinces in melon, cucumber and squash. When plant sap of ELISA positive samples was used to mechanically inoculate healthy squash plants, chlorotic spots with, in some cases, necrotic centres were observed on the inoculated leaves 20–25 days postinoculation. The presence of CLSV was confirmed by reverse transcription polymerase chain reactions using specific primers amplifying the entire coat protein gene of CLSV. Sequence comparison with sequences available at GenBank showed 93% nucleotide sequence identity to CLSV isolates from Israel (DQ227315) and Canada (EU127904), the only CLSV coat protein sequences available. To our knowledge, this is the first report of the occurrence of CLSV in Iran.

Eggplant (Solanum melongena), which is grown worldwide, is cultivated all year round in Israel. We report on the outbreak of a new viral disease infecting eggplants in Israel, which causes significant damage due to plant stunting, leaf mottling and narrowing, accompanied by fruit malformation. Virus purification from the infected plants yielded isometric particles, 37-40 nm in diameter. Biological and molecular characterization of the viral agent indicates that it is a new strain of the Eggplant mottled crinkle virus (EMCV-Is) that has been assigned to the Tombusvirus genus. Phylogenetic analysis indicated a close relationship of EMCV with Pear latent virus (PeLV) and Lisianthus necrosis virus (LNV), which can be considered strains of EMCV rather than distinct viruses.

Irrigation water can be polluted by the tobacco etch virus (TEV), which causes serious economic loss in tobacco. However, it is difficult to monitor the sanitary status of irrigation water because TEV presents at extremely low concentrations. This study designed a procedure for concentrating TEV from a standard water sample using polyethylene glycol and detecting it using SYBR Green-based quantitative real-time polymerase chain reaction (qRT-PCR). The concentration factors were optimized through orthogonal tests and the highest recovery efficiency of the TEV genome from a standard water sample was 92.05%. The sensitivity was evaluated using nine 10-fold dilution series of TEV plasmids, and the detection limit of the qRT-PCR was about 10 viral copies/µL. In the infectivity test, TEV was first detected in the roots of NC89 at 7 days after treatment and in the upper leaves at 14 days after treatment. Field diagnosis results showed that 56 of the 180 samples tested positive for TEV, which indicated that this method may be suitable for concentrating and detecting TEV in irrigation water.
