Characteristics of immune memory 10–15 years after primary hepatitis B vaccination

Abstract: The presence of B cell memory could be shown by a typical anamnestic anti-HBs response curve after a booster dose in all but one individual. In contrast, T cell responses to booster vaccination, which occurred in approximately 50% of participants, were rather heterogeneous.

“…Different definitions and timepoints have been used to measure the anamnestic response to a challenge dose. An HBsAg vaccine challenge has been shown to induce a typical anamnestic response with a steep increase starting between 3 and 7 days postchallenge, peaking around 14 days and plateauing between 14 days and 1 month . This is consistent with results from our study which show an initial increase in anti‐HBs concentrations 7 days postchallenge (with an 84.2% anamnestic response rate) and a further increase 30 days postchallenge.…”

“…Despite low frequencies of HBsAg‐specific CD4 + T cells in venous blood 20‐30 years after primary vaccination, the observed increase as early as 7 days after the HBsAg vaccine challenge indicates a memory recall. Previous studies have assessed long‐term persistence of memory B and CD4 + T cells and the cellular immune response to a challenge up to 32 years after priming in adults, infants or children . However, there was substantial heterogeneity in the assays and timepoints used for assessment; therefore, it is hard to compare results.…”

“…It is well established in literature that HBsAb can provide long-term protection in a general healthy population 20 22 . The protective role of HBsAb in clearance of circulating HBsAg and suppression of HBV replication was also reported.…”

“…Se conoce que la inmunidad contra el VHB es regulada por factores genéticos del hospedero, como las variantes alélicas de SNP de genes de la respuesta inmune y la expresión de ARNm de IFN-γ (4,9). La distribución de la frecuencia de alelos de IL-12A e IL-12B SNP fue similar a la identificada en los pacientes de Polonia y de Taiwán (10,11).…”
