Characteristics of live parameters of the HS‐5 human bone marrow stromal cell line cocultured with the leukemia cells in hypoxia, for the studies of leukemia–stroma cross‐talk

Abstract: The unique bone marrow microenvironment is created by stromal cells and such physical conditions as hypoxia. Both hypoxia and interactions with stromal cells have a significant impact on the biology of leukemia cells, changing their sensitivity to antileukemic therapies. Thus, it is crucial to introduce biological systems, which enable the investigation of leukemia-stroma cross-talk and verification of novel therapies effectiveness under such bone marrow niche-mimicking conditions. Here, we have established an… Show more

“…Although OP9 cells are excellent at supporting BCP-ALL cell proliferation and survival, some applications may need to utilize human MSC stromal support. We therefore tested the human MSC cell line HS-5, which has a similar pattern of gene expression to primary bone marrow MSC [ 29 ] and is widely used to support survival of malignant human hematopoietic cells [ 30 , 31 ]. As shown in Fig 7A , mitotically inactivated HS-5 cells plated alone have attached to the electrodes, causing an increase in cell index values over time.…”

“…Early relapse may arise when treatments are effective at eradicating leukemic cells in the peripheral blood but not in the bone marrow, where leukemic cells are sheltered. Thus, novel biological systems are currently established that enable the investigation of leukemia–stroma cross-talk and verification of novel therapies’ effectiveness under such bone marrow niche-mimicking conditions [ 27 ]. In our study, we employed a simple co-culture system with the HS-5 stroma cell line to grow AML cells in a bone-marrow niche environment.…”

“…Stromal cells were previously reported to provide chemoprotection to CML cells via secreted factors and direct cell-cell communication (Kolba et al, 2019;Kumar et al, 2017;Podszywalow-Bartnicka et al, 2018;Weisberg et al, 2008;Wolczyk et al, 2023). Various factors secreted by stromal cells, including inflammatory cytokines like IL-6 and IL-8, or TGFβ can protect CML cells from BCR-ABL1 kinase inhibition by kinase inhibitors such as imatinib (Civini et al, 2013;Le et al, 2020).…”
