Characteristics of thawed autologous umbilical cord blood

Abstract: The high variability and low counts found in AutoUCB banking suggest that further standardization of characterization, collection, and processing procedures is needed.

“…12 Many variables that may improve the quality of UCB are still under study and research, and these factors can lead to lowering the cost and time consumed in processing and storage of unsuitable UCB. 13,14 Many fetal and maternal physiologic factors such as birthweight, weight of placenta, sex of newborn, type of delivery and mode of collection are correlated with UCB volume and TNC count. 3,15 New donor selection strategies to improve the quantity and quality of UCB units must be proposed.…”

“…All samples were obtained under Institutional Review Board (IRB)-exempt approved protocols at UF (non-human exempt protocol IRB201300072). CryoCBUs were thawed as described previously, 53 resuspended in 60 mL of dextran-HSA wash solution (divided into two 50-mL conical tubes), allowed to warm to room temperature, and underlaid with 15 mL Ficoll-Paque PLUS per conical tube for density gradient centrifugation and CBMC isolation.…”

“…Recently, several structures have started programs of autologous CB (sometimes extended to the familial use for a family‐directed donation) private banking to store a stem cell source for a possible therapeutic use in unborn children. In this case, policies of banking with respect to TNC vary from bank to bank, depending on the country, on the specific agreement form which has been adopted at a given site and offered to the parents . In banks of unrelated CB, the count of TNCs in CBUs more than 80 to 60 mL is considered the best predictor of both CD34+ HSC content and successful engraftment after transplantation .…”
