Intestinal contents of suckling (n = 45) and newly weaned (n = 45) piglets, suffering from diarrhea in the province of Villa Clara in Cuba, were tested for viral, bacterial, and parasitic enteropathogens from May to June 2008. At least one enteropathogen was identified in 53.3 % of piglets and enterotoxigenic Escherichia coli (ETEC; 25.6 %) was the major pathogen; mostly STa(+)/STb(+) or F4(+)/STa(+)/STb(+) ETEC were isolated. The overall occurrence of the rest of pathogens was 10 % for transmissible gastroenteritis virus (TGEV) and Cryptosporidium parvum, 6.7 % for rotavirus A and Isospora suis, 5.6 % for α-toxigenic Clostridium perfringens, 3.3 % for verotoxigenic E. coli (VTEC), and 2.2 % for Salmonella enterica subspecies enterica serovar Newport. TGEV and α-toxigenic C. perfringens were only identified in suckling piglets, while Salmonella Newport and VTEC were only detected in weaned pigs. Porcine epidemic diarrhea virus (PEDV), β-toxigenic C. perfringens, Eimeria spp., and helminths were not identified. Eight kinds of mixed infections were detected in 25 % of enteropathogen positive piglets. ETEC was present in 10 of 12 mixed infections, and TGEV infections were never combined. This survey demonstrates that several enteropathogens are circulating in piggeries located in the province of Villa Clara in Cuba, and that is necessary to improve surveillance, prevention, and control of enteric infections in order to increase production efficiency.

La emergencia mundial de Salmonella multirresistente a antimicrobianos es de creciente preocupación, estudios recientes resaltan la relación entre el consumo de antibióticos por los animales y los diversos mecanismos de resistencia desarrollados por los microorganismos. En el presente estudio se investigó la circulación de Salmonella spp. asociada a procesos infecciosos en animales domésticos, así como los patrones de susceptibilidad antimicrobiana. El muestreo y el aislamiento de Salmonella spp. se llevaron a cabo en el Departamento de Bacteriología General del Laboratorio Provincial de Diagnóstico Veterinario de la provincia Villa Clara, Cuba, durante 2016. El método Kirby-Bauer se empleó para determinar la susceptibilidad antimicrobiana. Para el análisis de los resultados se empleó el paquete estadístico Statgraphics Centurion versión XV-II / 2006 aplicando procedimientos descriptivos, y pruebas de comparación múltiple de proporciones, para la evaluación de los porcentajes de resistencia de Salmonella por especie animal y por serogrupo. Se identificaron 46 aislados de Salmonella spp. pertenecientes a los serogrupos B, C1, C2 y D en el 15% de las muestras, todas provenientes de gallinas, bovinos, ovinos y cerdos. En general, los patrones de resistencia antimicrobiana fueron expresados para la ampicilina (en el 46% de los aislados), compuestos de sulfonamida (28%), ácido nalidíxico (26%) y tetraciclina (22%); no se detectó resistencia a ciprofloxacina, gentamicina y cefotaxima. Los aislados de Salmonella spp. más resistentes fueron de los serogrupos B y D, así como los provenientes de ovinos y cerdos.

The mortality of livestock caused by pathogenic Escherichia coli (E. coli) still accounts for a large proportion of deaths in large-scale production and reproduction, which causes devastating economic losses to the pig breeding industry. The aims of this study were to investigate the antibacterial activity of combined aditoprim (ADP) and sulfamethoxazole (SMZ) against clinical isolates of E. coli from pigs and to develop a pharmacokinetic-pharmacodynamic (PK-PD) model to formulate the optimal dose of ADP/SMZ for the treatment of pig colibacillosis. Blood and ileum fluid samples were collected at different times after single intramuscular injection of ADP/SMZ (5/25 mg/kg b.w.) to healthy pigs and E. coli-infected pigs. Concentrations of ADP and SMZ in plasma and ileum fluid were analyzed by HPLC. The peak concentration (C max ) and the area under the concentration-time curve (AUC 0-24h ) in ileum fluid of healthy pigs were 1.76 ± 0.27 µg/ml and 18.92 ± 2.87 µg•h/ml for ADP and 19.15 ± 2.63 µg/ ml and 125.70 ± 11.86 µg•h/ml for SMZ, respectively. C max and AUC 0-24h in ileum fluid of infected pigs were 1.88 ± 0.13 µg/ml and 15.12 ± 0.75 µg•h/ml for ADP and 19.71 ± 3.68 µg/ml and 133.92 ± 17.14 µg•h/ml for SMZ, respectively. The minimum inhibitory concentrations (MICs) of combined ADP and SMZ (ADP/SMZ) against 185 strains of E. coli from pigs were determined. The MIC 50 and MIC 90 of ADP/SMZ were 0.5/2.5 and 4/20 µg/ml, respectively. The MIC of the selected pathogenic E. coli SHC28 was 0.5/2.5 µg/ml in Mueller-Hinton broth and 0.25/1.25 µg/ml in ileum fluid, respectively. In vitro, the mutant prevention concentration, the post-antibiotic effect, growth, and time-killing curves in vitro and ex vivo of ADP/SMZ against the isolate SHC28 were assayed for PD studies. The results showed that ADP/SMZ exhibited strong concentration-dependent antimicrobial activity against E. coli.
