Characterization and Comparison of Clavibacter michiganensis subsp. nebraskensis Strains Recovered from Epiphytic and Symptomatic Infections of Maize in Iowa

Abstract: Clavibacter michiganensis subsp. nebraskensis (Cmn), the causal organism of Goss’s wilt and leaf blight of maize, can be detected in the phyllosphere of its host prior to disease development. We compared the morphology and pathogenicity of 37 putative isolates of Cmn recovered from asymptomatic and symptomatic maize leaves. Thirty-three of the isolates produced mucoid orange colonies, irrespective of the source of isolation and all but four of these isolates were pathogenic on maize. The remaining 4 isolates r… Show more

“…V3 corn developmental stage plants were used for the inoculation following a slightly modified dropped‐wounded protocol described by Ahmad et al . (). The third leaf of each plant was wounded halfway between the sheath and the tip by making a cut across three veins on one side of the midrib.…”

“…C. michiganensis ssp. nebraskensis strain 2581 isolated from maize does not carry any plasmids (Eichenlaub & Gartemann, 2011;Ahmad et al, 2015), and its putative virulence genes are located in the chromosome, although their identity and function have not been well studied (Tambong, 2017). C. michiganensis ssp.…”

“…For single gene dnaA, the best evolution model was determined, and maximum likelihood (ML) method with 1,000 bootstrap values to evaluate node support was used to infer the phylogenetic tree. A color-coded matrix showing pairwise identity was generated using Sequence Demarcation Tool version 1.2 [23].…”
