Characterization and cost–benefit analysis of automated bioreactor‐expanded mesenchymal stem cells for clinical applications

Abstract: MSCs manufactured by an automated bioreactor are physically and functionally equivalent to the MSCs manufactured by the manual flask method and have met the standards required for clinical application.

“…Bioreactors are an effective and scalable strategy for MSC expansion compared to traditional monolayer culture (7,22,23). Beyond expansion, perfusion flow-induced shear stress promotes an osteogenic phenotype (24) and deposition of a mineralized ECM by MSCs (25,26).…”

“…Particularly for MSCs, it is important to consider from the outset how amendable these new methods would be for different types of scalable MSC manufacture approaches. For example, dynamic-culture bioreactors or planar-culture 'cell factories' [91][92][93][94], which may ultimately dictate the method employed. So far, the senescence status of MSCs grown on microcarriers in bioreactors continues to be assessed off-line [22] following MSC harvesting, plating in 2D and SA--gal activity staining in monolayer conditions [95].…”

“…Developers of laboratory of automation have attempted to minimise the footprint of their machinery through innovative reworkings of traditional laboratory procedures. The use of hollow fibre arrays (Russell et al, 2018) and multiaxis liquid and labware manipulation (Kato et al, 2010) are examples of compact automated adherent cell culture systems. Spatial constraints may push future bench-based laboratory automation towards an architectural style resembling inner city skyscrapers.…”
