Characterization and overexpression of a novel keratinase from Bacillus polyfermenticus B4 in recombinant Bacillus subtilis

Abstract: Background: Keratins, insoluble proteins with a robust structure, are a major component of epidermal tissue and appendages such as hair, feathers, nails, and walls. Keratinous waste mainly emanates from poultry and leather industries, thereby severely contaminating the environment. Keratinase can lyse proteins with robust cross-linked structures, such as keratin, and can hence be used in animal feed, fertilizer, detergent, leather, pharmaceutical, and cosmetic industries. Bacillus polyfermenticus B4, isolated … Show more

“…Pressurized cooking or chemical treatment of feathers was the fundamental method of processing used in preparing feather meal. This method of processing involves high energy input, and it, therefore, denatures important heatlabile proteins (Onifade et al, 1998;Dong et al, 2017). The potentials presented by keratinolytic microbes and keratinases are an indication of the possibilities that the bio-recycling process holds for the husbandry sector of the agro-industry (Gegeckas et al, 2018).…”

“…Keratin degradation is attracting biotechnological attention since it might provide a substitute way of waste management as well as the production of valuable products (Brandeli andRiffel 2006, Pasupuleti et al, 2010). Bacillus was used for their keratinases production, optimization and characterization (Sahoo et al, 2012, Dong et al, 2017, Arokiyaraj et al, 2019. Tiwary and Gupta (2012) used Bacillus licheniformis to study the conversion of the chicken feather into feather meal.…”

“…Diverse groups of microorganisms are reported to produce keratinases (Kalia and Purohit, 2008 ), such as numerous Bacillus sp. (Gupta and Singh, 2013 ; Gupta et al, 2015 ; Dong et al, 2017 ; Hamiche et al, 2019 ; Nnolim et al, 2020b ; Almahasheer et al, 2022 ; Devi et al, 2022 ; Mazotto et al, 2022 ), Aspergillus oryzae (Farag and Hassan, 2004 ), Paecilomyces marquandii and Doratomyces microsporum (Gradisar et al, 2005 ), Streptomyces sp. (Tatineni et al, 2008 ), Stenotrophomonas maltophilia (Cao et al, 2009 ), Myrothecium verrucaria (Moreira-Gasparin et al, 2009 ), Thermoactinomyces sp.…”
