Characterization of 18 new microsatellite loci in Atlantic cod (Gadus morhua L.)

Abstract: Eighteen new microsatellite loci consisting of 10 di-, 5 tri-, 2 tetra- and 1 heptanucleotide repeats are introduced for the Atlantic cod (Gadus morhua L.). All loci were co-amplified in two polymerase chain reactions (plus two previously published microsatellites) and all products were typed clearly. The number of alleles per locus ranged from six (PGmo130) to 45 (PGmo76) and the observed heterozygosity ranged from 0.356 (PGmo130) to 0.957 (PGmo95). All loci except one followed Hardy-Weinberg expectations. Ge… Show more

“…From this sample, 757 individuals were assigned to 40 families that are 40 females and 20 males. Genotyping was determined from the following 16 microsatellites: Gmo8, Gmo19, Gmo37 (Miller, Le & Beacham ), Tch11, Tch14 (O'Reilly, Canino, Bailey & Bentzen ), Gmo38 (Jakobsdottir, Jorundsdottir, Skirnisdottir, Hjorleifsdottir, Hreggvidsson, Danielsdottir & Pampoulie ), PGmo38, PGmo49, PGmo61‐FRb, PGmo71, PGmo74, PGmo87, PGmo94, PGmo100, PGmo124 and PGmo134 (Skirnisdottir, Pampoulie, Hauksdottir, Schulte, Olafsson, Hreggvidsson & Hjorleifsdottir ).…”

“…To attain the goal of 20 microsatellite loci, 19 walleye pollock loci (O'Reilly et al 2000), 19 Atlantic cod loci (Miller et al 2000;Stenvik et al 2006;Wesmajervi et al 2007;Skirinsdottir et al 2008), and 11 Pacific cod loci (Cunningham et al 2009) were screened. One additional locus (Gma107), which had not been used previously (Canino et al 2005), was also evaluated.…”

“…Samples were genotyped at 18 polymorphic microsatellite loci: Gmo2 (Brooker et al 1994), Gmo8, Gmo19, Gmo34, Gmo37 (Miller et al 2000), Tch5, Tch11, Tch14, Tch22 (O'Reilly et al 2000, PGmo38, PGmo49 (Jakobsdóttir et al 2006), andPGmo61, PGmo74, PGmo87, PGmo94, PGmo100, PGmo124, andPGmo134 (Skirnisdottir et al 2008). Polymerase chain reaction (PCR) conditions followed the method of Pampoulie et al (2006) for the first set of markers (Gmo8,Gmo19,Gmo34,Gmo37,Tch5,Tch11,Tch14,and Tch22) and of Skirnisdottir et al (2008) for the second set (PGmo38,PGmo49,PGmo61,PGmo74,PGmo87,PGmo94,PGmo100,PGmo124,and PGmo134). Samples were then analyzed on an ABI PRISM 3730 sequencer with the GeneScan-500 LIZ size standard and genotyping was performed with GeneMapper version 3.5 (Applied Biosystems, Foster City, California).…”
