Characterization of a membrane protein (VP001L) from infectious spleen and kidney necrosis virus (ISKNV)

Abstract: Infectious spleen and kidney necrosis virus (ISKNV) is the type species of megalocytivirus, Iridoviridae. A novel membrane protein corresponding to the first open reading frame (ORF001L) of ISKNV genome was identified. This 378-residue protein, termed the VP001L protein, has a high content of hydrophobic sequences and contains 10-11 putative transmembrane domains, indicating it may be a membrane protein. The VP001L mRNA start site was extended 433 bp upstream of the start codon and the temporal analysis showed… Show more

“…ORF1L exhibited high homology to the amino acid sequences of ORF1L of ISKNV and OSGIV [14,15]. ORF1L of ISKNV, containing 10 ~ 11 putative transmembrane domains, is a membrane protein and is considered to serve as a model for analyzing the topology and roles of different hydrophobic regions in multi-transmembrane proteins [36]. …”

“…Further research is required to resolve this question. Recently, a number of studies have been undertaken to identify gene function in ISKNV [47,48,49,50,51]. These expanding biochemical studies will clarify the function of specific ISKNV genes and indicate whether megalocytiviruses display replicative strategies unique to this viral genus.…”

“…To better understand the molecular mechanism of iridovirus pathogenesis and explore the strategy for prevention and control iridovirus diseases, complete genomes of 19 iridovirus isolates were sequenced and annotated up to now [6-8]. Some functional genes encoded by individual iridovirus, such as infection cell protein (ICP), capsid protein, membrane protein and virus replication associated enzymes have been identified and characterized [9-11]. However, the function of many iridovirus core structural genes remained largely unknown.…”
