Characterization of a novel mycotombus-like virus from the plant-pathogenic fungus Phoma matteucciicola

Abstract: Here, we report a novel mycotombus-like mycovirus, tentatively named 'Phoma matteucciicola RNA virus 2' (PmRV2), derived from the phytopathogenic fungus Phoma matteucciicola strain HNQH1. The complete PmRV2 genome is comprised of 3,460 nucleotides (nts), a positive-sense single-stranded RNA (+ ssRNA) with a GC content of 56.71%. Sequence analysis of PmRV2 indicated the presence of two noncontiguous open reading frames (ORFs) encoding a hypothetical protein and a RNA-dependent RNA polymerase (RdRp), respectivel… Show more

“…With global travel and trade, caution has to be taken for a potential Clade I MPXV outbreak which is more virulent than Clade II MPXV. Rapid and accurate diagnostic methods are the foundation of effective prevention and control of mpox outbreaks 25 . In addition to antigen/antibody‐based serology tests, PCR‐based diagnosis has also becoming widely adopted as a result of its specificity and high sensitivity 31,32 .…”

“…RAA‐coupled lateral flow strips allow the simplest detection procedure with the same sensitivity. Moreover, the biggest impediment for field adaptation is sample preparation, which is often a multistep procedure requiring columns or magnetic beads and depends on laboratory instruments 24,25 . It is believed that one‐step sample preparation and fast detection technology together enable rapid, point‐of‐care detection of MPXV, allow early identification and self‐isolation of mpox.…”

“…Polymerase chain reaction (PCR), reverse‐transcription polymerase chain reaction (RT‐PCR), isothermal amplification (loop‐mediated amplification & recombinase polymerase amplification), CRISPR, immunological methods (ELISA detection, haemagglutination inhibition test, western blot, and radioimmunoassay), whole genome sequencing and MPV isolation and culture techniques are the identified methods still now for MPV detection 28 . Among the diagnostic methods of MPV, RT‐PCR has been one of the most effective techniques till now due to its higher sensitivity, specificity, rapidity, and accurate outcomes.…”

“…However, diagnosis of mpox is difficult. Confirmation of mpox diagnosis is primarily by PCR, (4, 5) but, concerningly, new evidence suggests that the Clade I-specific RT-PCR test recommended by the US CDC is impacted due to genetic mutations in the virus causing this outbreak. (6) There are no point-of-care or rapid diagnostic tests alternatives at present.…”
