Characterization of aldosterone binding sites in circulating human mononuclear leukocytes

Abstract: Aldosterone binding sites in human mononuclear leukocytes were characterized after separation of cells from blood by a Percoll gradient. After washing and resuspension in RPMI-1640 medium, cells were incubated at 37 degrees C for 1 h with different concentrations of [3H]aldosterone plus a 100-fold concentration of RU-26988 (11 alpha, 17 alpha-dihydroxy-17 beta-propynylandrost-1,4,6-trien-3-one), with or without an excess of unlabeled aldosterone. Aldosterone binds to a single class of receptors with an affinit… Show more

“…As mentioned above, MR are also present in non-epithelial cells/ tissues such as the hippocampus (McEwen & Wallach 1973), macrophages (Armanini et al 1985a, Lim et al 2007, VSMC, placenta and heart (Pearce & Funder 1988, Lombes et al 1992. In the absence of the aldosterone specificity-conferring enzyme 11bHSD2, MR are overwhelmingly occupied by glucocorticoids.…”

“…ALDO binds to a single class of cytosolic receptors in these cells. 37 In patients with either PAL or renin-dependent, secondary aldosteronism (SAL),ALDO binding sites are reduced by 50%; following surgical removal of adenomatous adrenal tissue, receptor binding in PBMC is normalised. 38,39 ALDO promotes the efflux of Mg 2+ from cultured human lymphocytes in exchange for Na + via receptor-ligand binding.…”

“…Several investigators reported that not only Ang II but also aldosterone induced the activation of nuclear factor (NF)-ÎșB through either AT1R (9,11,24,25) or MR (15,26). NF-ÎșB enhances gene expressions involved in inflammation with subsequent cytokine production such as monocyte chemoattractant protein-1 (MCP-1) (27) and tumor necrosis factor-α (TNF-α) (11) in human PBMCs.…”
