“…These I Ks activators work from outside the cell membrane by shifting the voltage dependence of Q1 activation in the negative direction. The degree of shift is enhanced by E1 association ( Wu et al, 2006b ). Mutations or deletions in the 39 -42 region of E1 abolish these activators ' actions ( Abitbol et al, 1999 ).…”

“…E1 makes extensive contacts with Q1, including the transmembrane region (Melman et al 2001; Panaghie et al 2006; Tai and Goldstein 1998; Tapper and George 2001), the cytoplasmic region (Haitin et al 2009; Lvov et al 2010; Wiener et al 2008), and the extracellular region (Chung et al 2009; Nakajo and Kubo 2007; Xu et al 2008). Q1/E1 interactions in the transmembrane region determine the I Ks pore conductance (Sesti and Goldstein 1998; Yang and Sigworth 1998) and gating kinetics (Melman et al 2001), while Q1/E1 interactions in the cytoplasmic region influence I Ks trafficking (Ghosh et al 2006; Shamgar et al 2006), modulation by protein kinase A-mediated phosphorylation (Kurokawa et al 2009), and gating kinetics (Lvov et al 2010; Wu et al 2006). However, the functional roles of Q1/E1 interactions in the extracellular region are not clear.…”

“…Interestingly, KCNE1, CACNA1C, and KCNQ2 are already targets of drugs against arrhythmia, atrial fibrillation, congestive heart failure, left ventricular hypertrophy, and isolated systolic hypertension (42). Furthermore, there is recent evidence that the post-transmembrane domain region of KCNE1 interacts with the KCNQ1 channel to modulate the I(K) current amplitude and gating kinetics (43).…”
