Characterization of Anthocyanins in Grape Juices by Ion Trap Liquid Chromatography−Mass Spectrometry

Abstract: A reverse phase HPLC and electrospray interface with ion trap mass spectrometer method was developed for the characterization of anthocyanins in Concord, Rubired, and Salvador grape juices. Rubired and Salvador grapes are hybrids from Vitis vinifera and Vitis rupestris. Concord grape is a grape from the native American cultivar Vitis labrusca. Individual anthocyanins in these three varieties were identified on the basis of UV-vis and MS spectra and further elucidated by MS/MS spectra. Anthocyanins in Salvador … Show more

“…The [M+H] + ion at m/z 479*(* stands precursor ions generated from ESI+ mode) was detected in both RGPE and WGPE (Figure 1 C,D), which was assigned to petunidin-3-glucoside confirmed by its fragments at m/z 303 and 317 (Table 2) [26]. The [M+H] + ions at m/z 463*, 493*, 505*, 535*, 625*, 639*, and 655* could be assigned to peonidin 3-glucoside, malvidin 3-glucoside, peonidin 3-acetylglucoside, malvidin 3-acetylglucoside, petunidin 3-p-coumaroylglucoside, malvidin 3-p-coumaroylglucoside, and malvidin 3-(6-O-caffeoyl) monoglucoside (Figure 1 C,D (Table 2), respectively [26][27][28]. Of which, peonidin 3-glucoside and peonidin 3-acetylglucoside were detected in both RGPE and WGPE, while malvidin derivatives and petunidin 3-p-coumaroylgluside only found in RGPE ( Table 2).…”

“…A phenolic wine extract was prepared according to previously described methodologies [22,23] with some changes. Briefly, dealcoholized wine was applied to a column Sep-Pak C 18 , 10 g (Waters) previously conditioned with methanol 5 ml/min for 20 min followed by water:formic acid (95:5) 5 ml/min for additional 20 min.…”

“…The identification of five basic non-acylated anthocyanins was confirmed by comparison with commercially available anthocyanin standards and the MS/MS spectra. Identification of all the other acylated anthocyanins was achieved mainly by comparing molecular ions and product ions of these anthocyanins with those available in the literature (García-Beneytez et al, 2002Wang et al, 2003;Núñez et al, 2004;De Villiers et al, 2004;Downey & Rochfort, 2008;Han et al, 2008;Cook Papini et al, 2010). The cis and trans isomers of the coumaroylates for peonidin-3-Oglucoside and malvidin-3-O-glucoside were distinguished by elution time and proportion.…”
