Characterization of B‐cell subpopulations in patients with chronic rhinosinusitis

Abstract: This study provides further evidence of an important role of B cells in CRSwNP patients. The local increase appears to be independent of a systemic response.

“…Because so many cell types, including most immune cells and epithelial cells, express 1α‐hydroxylase, we elected to examine total sinonasal 1α‐hydroxylase in this study. This was largely because although we know epithelial cell–specific 1α‐hydroxylase is reduced in CRSwNP, the number of sinonasal immune cells are increased in CRSwNP and AFRS . Therefore, it was possible that the increased immune infiltrate could impact total 1α‐hydroxylase levels.…”

“…HSNECs are one of many cell types found in the sinonasal mucosa that express 1α‐hydroxylase, in addition to monocytes, macrophages, B‐cells, and dendritic cells . Given that CRSwNP patients have elevated sinonasal levels of each of these immune cell populations, it is possible that total 1α‐hydroxylase could be increased or unchanged locally. Furthermore, it remains to be determined if sinonasal reductions in 1α‐hydroxylase are associated with reductions in 1,25VD3, which we examined in these studies.…”

“…Hair samples were analyzed for nicotine as described in an earlier study . Sinonasal tissue was collected, processed, and stored as described previously . Blood from control patients was processed as described previously, to obtain peripheral blood mononuclear cells (PBMCs), which were cryogenically preserved for later use …”
