Characterization of bifunctional l-glutathione synthetases from Actinobacillus pleuropneumoniae and Actinobacillus succinogenes for efficient glutathione biosynthesis

Abstract: Glutathione (GSH), an important bioactive substance, is widely applied in pharmaceutical and food industries. In this work, two bifunctional L-glutathione synthetases (GshF) from Actinobacillus pleuropneumoniae (GshFAp) and Actinobacillus succinogenes (GshFAs) were successfully expressed in Escherichia coli BL-21(DE3). Similar to the GshF from Streptococcus thermophilus (GshFSt), GshFAp and GshFAs can be applied for high titer GSH production because they are less sensitive to end-product inhibition (Ki values … Show more

“…GSH is produced through two sequential enzymatic reactions catalyzed by glutamate-cysteine ligase ( gsh A) and GSH synthetase ( gsh B). In some bacteria, GSH is produced by a bifunctional enzyme (GshF) catalyzing both reactions simultaneously (Yang et al, 2016). Prokaryotes characterized by the absence of GSH, produce other LMW thiols – such as mycothiol, bacillithiol and CoA – with a similar protective role to GSH (Fahey, 2013).…”

“…It was confirmed that GshFSS exhibited the highest specific activity (841 U/mg). Interestingly, although GshFSS was not derived from a thermophilic microorganism, its activity at 45°C was higher than that of GshFST (Yang et al, 2016). This result showed its potential application for GSH synthesis.…”

“…One unit of enzyme activity was defined as the amount of enzyme that produces 1 mM GSH per hour (Yang et al, 2016). All the measurements were performed in triplicates.…”
