The Botryosphaeriaceae represents an important and diverse family of latent fungal pathogens of woody plants. While some species appear to have wide host ranges, others are reported only from single hosts. It is, however, not clear whether apparently narrow host ranges reflect specificity or if this is an artifact of sampling. We address this question by sampling leaves and branches of native South African trees from four different families, including Acacia karroo (Fabaceae), Celtis africana (Cannabaceae), Searsia lancea (Anacardiaceae) and Gymnosporia buxifolia (Celastraceae). As part of this process, two new species of the Botryosphaeriaceae, namely Tiarosporella africana sp. nov. and Aplosporella javeedii sp. nov., emerged from sequence comparisons based on the ITS rDNA, TEF-1α, β-tubulin and LSU rDNA gene regions. An additional five known species were identified including Neofusicoccum parvum, N. kwambonambiense, Spencermartinsia viticola, Diplodia pseudoseriata and Botryosphaeria dothidea. Despite extensive sampling of the trees, some of these species were not isolated on many of the hosts as was expected. For example, B. dothidea, which is known to have a broad host range but was found only on A. karroo. This could have resulted from the fact that it is a rare species in the region. With the exception of S. lancea, which was infected by A. javeedii all the hosts were infected by more than one Botryosphaeriaceae species. Collectively, the results suggest that some intrinsic host factors, possibly combined with local environmental conditions, affect the distribution and co-infectivity of various hosts by the Botryosphaeriaceae. 2This would counteract the general ability of a species in the Botryosphaeriaceae to infect a broad range of plants. The combination of host and environmental factors might also explain why some Botryosphaeriaceae with apparently broad host ranges, are found on different suites of hosts in different areas of the world.

We have explored the fungal diversity in asymptomatic twigs of apple, peach, pear and blueberry trees, with the objective of discerning between true endophytes and latent pathogens. Several fungal genera containing known bark pathogens were found. Seven Diaporthe species—D. oxe, D. infecunda, D. serafiniae, D. phaseolorum, D. terebinthifolii, D. foeniculina and D. brasiliensis—were identified, along with Botryosphaeria dothidea, Neofusicoccum parvum, Neofusicoccum australe, Cytospora sp., Cytospora acaciae and Pestalotiopsis spp. A pathogenicity trial was undertaken to determine the role of these species on apple, pear, blueberry and peach shoots. Diaporthe brasiliensis, D. foeniculina, Diaporthe inconspicua, D. terebinthifolii, Diaporthe sp.1, Cytospora‐like isolates and Pestalotiopsis spp. isolates produced no lesions on inoculated shoots, suggesting that they could be considered true endophytes on their respective hosts. Meanwhile, some of the isolates of Diaporthe—D. oxe, Diaporthe sp.2, D. infecunda and D. serafiniae, B. dothidea, N. parvum and N. australe could be regarded as latent pathogens in their respective hosts as they produced sunken cankers and necrosis on inoculated shoots. These results demonstrate that apple, pear, blueberry and peach healthy shoots can host many known endophytic fungi along with potential wood disease‐causing fungi that should be regarded as latent pathogens.

N. kwambonambiense was described for the first time in South Africa in asymptomatic stems, leaves and fruits of dead plants of Syzygium cordatum (Pavilic et al 2009, Phillips et al 2013 and was later reported in grapevines in Uruguay (Abreo et al 2013), and to cause fruit rot in post-harvested strawberry in Brazil (Lopes et al 2014). The occurrence of N. kwambonambiense associated with symptoms of dieback and canker in stems of cashew plants is unprecedented in Brazilian territory.…”

The Botryosphaeriaceae family is one of the most important groups of plant pathogenic fungi associated with tropical woody species such as Anacardium occidentale (cashew) and Mangifera indica (mango), which are fruiting species with broad distributions and high economic importance in north-eastern Brazil. This study was designed to characterize species of this family associated with cashew, mango and Psidium guajava (guava) that cause dieback and stem and branch cankers. Characterization comprised phylogenetic, morphological, physiological and pathogenic features. The phylogenetic study combined sequences of the internal transcribed spacer (ITS) of the rDNA and partial elongation factor 1-α (EF1-α) regions of DNA. Measurements of fungal conidia and colony growth on different culture media and at different temperatures were conducted. Pathogenicity tests were also performed through inoculation of different host species. By combining the sequences of the referenced genes, it was possible to identify the following Botryosphaeriaceae species: Neofusicoccum kwambonambiense, Neoscytalidium hyalinum and Pseudofusicoccum stromaticum as the first report in cashew, Neoscytalidium hyalinum in mango and Neofusicoccum brasiliense as the first report in guava. Examination of N. brasiliense revealed conidia, providing the first morphological description of the species. Neofusicoccum kwambonambiense did not sporulate, but the other isolates produced conidia with dimensions comparable to those described in the literature. All isolates were virulent to mango fruits and young cashew and “caja-umbu” plants, causing necrotic lesions and gum exudation by the hosts. The isolates of Neofusicoccum were the most aggressive in the inoculated hosts.
