Characterization of cloned rat liver Na(+)-bile acid cotransporter using peptide and fusion protein antibodies

Abstract: A cDNA encoding a rat liver Na(+)-bile acid cotransporter (Ntcp) has recently been cloned (Hagenbuch, B., B. Steiger, M. Fouget, H. Lubbert, and P. J. Meier. Proc. Natl. Acad. Sci. USA 88: 10629, 1991) using expression cloning in Xenopus laevis oocytes. Although the open reading frame coded for a protein of 39 kDa, in vitro translation experiments produced a 35-kDa protein which increased to a product of 41 kDa after glycosylation by pancreatic microsomes. To more clearly characterize the native protein in rat… Show more

“…Two different proteins, namely Na ϩ /TC cotransport polypeptide (Ntcp) and epoxy hydrolase, have been shown to mediate Na ϩ /TC cotransport into hepatocytes (3, 4). The rat liver Ntcp is a ϳ51-kDa glycoprotein with seven transmembrane domains (3,5,6). Recent studies suggest that Na ϩ /TC cotransport and Ntcp are up-regulated by hormones and down-regulated by cholestatic agents.…”

“…17,26 We examined the cellular localization of ntcp in WIF-B cells grown for 10 days in the absence or presence of TC (50 µmol/L). Cells were fixed, permeabilized, and immunostained with a polyclonal antiserum directed against ntcp, as described in Materials and Methods.…”
