Characterization of diazotrophic root endophytes in Chinese silvergrass (Miscanthus sinensis)

Abstract: Background Phytoremediation is a potentially cost-effective way to remediate highly contaminated mine tailing sites. However, nutrient limitations, especially the deficiency of nitrogen (N), can hinder the growth of plants and impair the phytoremediation of mine tailings. Nevertheless, pioneer plants can successfully colonize mine tailings and exhibit potential for tailing phytoremediation. Diazotrophs, especially diazotrophic endophytes, can promote the growth of their host plants. This was te… Show more

“…Pseudomonas has been shown to suppress plant pathogens and promote plant growth ( Tao et al., 2020 ). In addition, Pseudomonas has a strong N fixation capacity ( Li et al., 2022b ) and can provide N for P.communis , which may also be an important reason why P.communis can adapt to N-deprived habitat. Both the Cyclobacteriaceae family and MND1 genera were also found to promote plant growth ( Li et al., 2020 ; Cheng et al., 2022 ), which is consistent with our study.…”

“…The Sb-dependent BNF activity assay includes nitrogenase activity measurements and Sb speciation (Sb(V) and Sb(III)) analysis. Acetylene reduction assay (ARA) was applied to analyze nitrogenase activity. Briefly, 60 mL sterile bottles were filled with 30 mL slurries of 1 g soil or tailing samples. Half of the experimental bottles were supplemented with 1 mM Sb(III) and 3 mM NaHCO 3 (Sb treatment) or 3 mM NaHCO 3 as a control treatment (No-Sb control).…”
