Characterization of DNA Binding Property of the HIV-1 Host Factor and Tumor Suppressor Protein Integrase Interactor 1 (INI1/hSNF5)

Abstract: Integrase Interactor 1 (INI1/hSNF5) is a component of the hSWI/SNF chromatin remodeling complex. The INI1 gene is either deleted or mutated in rhabdoid cancers like ATRT (Atypical terratoid and rhabdoid tumor). INI1 is also a host factor for HIV-1 replication. INI1 binds DNA non-specifically. However, the mechanism of DNA binding and its biological role are unknown. From agarose gel retardation assay (AGRA), Ni-NTA pull-down and atomic force microscopy (AFM) studies we show that amino acids 105–183 of INI1 com… Show more

“…We next investigated whether the observed alteration in conformation/antigenicity of 4-2.J41ΔCT was due to inefficient cleavage of the ΔCT version as uncleaved Envs bind to non-NAbs [ 8 , 20 , 21 , 33 ]. We carried out neutravidin-agarose pull down of biotinylated cell surface proteins from 4-2.J41 WT and 4-2.J41ΔCT 711 transfected cells and then probed the precipitate in western blots with anti-clade C antibodies (Fig.…”

“…At amino acid 7, there is a T/A heterozygous mutation (Glutamic acid/Valine) leading to sickle cell trait. ( d ) Mapping of how exons of SMARCB1 correlate to known domains of SMARCB1 (Allen et al, 2015; Das et al, 2013). ( e ) Quantitative reverse transcriptase PCR (qRT-PCR) of each exon-exon junction of SMARCB1 confirms expression loss in RMC samples.…”
