“…PBMCs were prepared by Ficoll gradient centrifugation (Lymphoprep, Axis-Shield, Dundee, Scotland), as described previously (32), from buffy coats of human blood from healthy donors provided by the blood bank of the University of Bonn. Purification of PDCs was accomplished by a combination of negative and positive selection with immunomagnetic beads.…”

“…Intracellular staining was performed as reported in detail elsewhere (32). MAbs used included PE-Cy5-labeled mAb against CD123 (9F5), PE-labeled mAb against IL-1b (8516.311), PE-labeled mAb against IL-6 (AS12), PE-labeled mAb against TNF-a (MAb11), and PE-labeled mouse IgG1k isotype control (MOPC-21), all purchased from BD Biosciences (Heidelberg, Germany).…”

“…Two major types of DC have been characterized in humans: the CD11c ϩ CD123 Ϫ myeloid (mDC, DC1), for which there are several subtypes, and the CD11c Ϫ CD123 ϩ lymphoid (DC2), also known as the plasmacytoid DC (pDC) (1). Studies have long supported the presence of the high-affinity IgE receptor (Fc⑀RI) on mDC, particularly those from allergic subjects, but have only recently identified this receptor on pDC (2,3). However, unlike the classical ␣␤␥ 2 tetramer that is uniquely expressed on basophils and mast cells and is involved in triggering mediator release from these cells, the role of the ␣␥ 2 variant on mDC and pDC is less clear, although the data at this time support it having a role in allergen presentation or "focusing" (4).…”

“…It has been reported that low numbers of plasmacytoid DCs in the epidermal skin lesions of AD patients allows them to be highly prone to viral skin infections like herpes simplex-induced eczema herpeticum. 83 …”
