Characterization of five marine family 29 glycoside hydrolases reveals an α-L-fucosidase targeting specifically Fuc(α1,4)GlcNAc

Abstract: l-Fucose is the most widely distributed l-hexose in marine and terrestrial environments, and presents a variety of functional roles. l-Fucose is the major monosaccharide in the polysaccharide fucoidan from cell walls of brown algae, and is found in human milk oligosaccharides and the Lewis blood group system, where it is important in cell signaling and immune response stimulation. Removal of fucose from these biomolecules is catalyzed by fucosidases belonging to different carbohydrate-active enzyme (CAZy) fami… Show more

“…Ri GH29 2A belonging to cluster 2 showed preference for α1,2 Gal linkages and displayed transfucosylation activity, as also reported for FgFCO1 from Fusarium graminearum PH-1 belonging to the same cluster 14 , 35 , 56 , 57 . La GH29 3A and Rs GH29 3A belonging to cluster 3 showed highest activity towards pNP-Fuc but no significant activity was found towards α1,2/3/4/6 fucosylated substrates, as also reported for Fp251, Fp239 and Fp231 from Paraglaciecola sp 58 , Alf1_Wf from W. fucaniytia CZ1127 T38 and ATCC_03833 3A from R. gnavus ATCC 29149 11 found in the same cluster. Ny GH29 4A belonging to cluster 4 showed preference for Fucα1,2 Gal, consistent with Fucosidase O from Omnitrophica bacterium OLB16 59 , HsFucA1 9 and Hs FucA2 60 from Homo sapiens found in the same cluster.…”

“…Some functionally characterised bacterial GH29 fucosidases have only been reported to be active against artificial substrates, such as BF0810 from Bateroides fragilis NCTC 9343 [ 56 ], Fp240 and Fp251 from Paraglaciecola sp. [ 57 ]. Further investigation is required to determine their specificity towards natural substrates.…”
