Characterization of hydrophobic-ligand-binding proteins of Taenia solium that are expressed specifically in the adult stage

Abstract: Taenia solium, a causative agent of taeniasis and cysticercosis, has evolved a repertoire of lipid uptake mechanisms. Proteome analysis of T. solium excretory-secretory products (TsESP) identified 10 kDa proteins displaying significant sequence identity with cestode hydrophobic-ligand-binding-proteins (HLBPs). Two distinct 362- and 352-bp-long cDNAs encoding 264- and 258-bp-long open reading frames (87 and 85 amino acid polypeptides) were isolated by mining the T. solium expressed sequence tags and a cDNA libr… Show more

“…Interestingly, we detected Eg HLBP in HF, while both Ts HLBP1 and Ts HLBP2 were not detected by Western Blot in the metacestode and showed a high expression in the T . solium adult [44]. In addition, we identified the Lipid transport protein N-terminal in both sQS f and bQS f .…”

“…Thirty fresh worms were incubated in 25 ml RPMI 1640 (Gibco) supplemented with the protease inhibitor cocktail for 1 h at 37°C. Addition of protease inhibitor cocktail into culture medium did not induce harmful effects on the excretory-secretory products (ESP) [28]. The incubation medium was harvested and centrifuged at 500 g for 10 min followed by 20000 g for 1 h. The supernatants were used as ESP.…”

“…HLBPs in T. solium are polymeric molecules consisting of protein subunits encoded by two gene families. One gene family encodes subunits with a molecular weight of 7 kDa ( RS1, Ts14, Ts18 ), whereas the other gene family encodes 10 kDa subunits ( CyDA, b1, m13h, TsHLBP1, TsHLBP2 ) (Kim et al, 2011; Rahman et al, 2012). These subunits preferentially bind different hydrophobic ligands, which is likely a reflection of their diverse primary structure and/or differential adaptation.…”
