Characterization of Arabidopsis sterol glycosyltransferase TTG15/UGT80B1 role during freeze and heat stress

Abstract: Sterol glycosyltransferases regulate the properties of sterols by catalyzing the transfer of carbohydrate molecules to the sterol moiety for the synthesis of steryl glycosides and acyl steryl glycosides. We have analyzed the functional role of TTG15/UGT80B1 gene of Arabidopsis thaliana in freeze/thaw and heat shock stress using T-DNA insertional sgt knockout mutants. Quantitative study of spatial as well as temporal gene expression showed tissue-specific and dynamic expression patterns throughout the growth st… Show more

“…For example, overexpression of cis‐ vaccenic acid in transgenic tomato ( Solanum lycopersicum ) lines resulted in an improved tolerance to freezing temperatures (De Palma et al ., ; Badea and Basu, ). Moreover, the apple samples treated with exogenous application of 1‐MCP showed a higher concentration of desmosterol, a sterol that confers adaptation to freezing stress in A. thaliana (Mishra et al ., ). The other apple samples in this current study (the two controls and T2_1‐MCP) were more oriented towards the projection of other classes of lipids in the PCA plot.…”

“…For salt and osmotic/drought stresses, 100 mM NaCl and 20% polyethylene glycol (PEG) was supplied, respectively ( Srivastava et al, 2012 ; Nautiyal et al, 2013 ). For desiccation, seedlings were transferred to sterile Whatman filter paper and for heat, cold and freeze stresses, rice seedlings were transferred to 45°C, ∼4°C, and ≤0°C, respectively ( Mishra et al, 2015 ). For various phytohormone treatments, both inoculated and uninoculated seedlings 100 μm each of ABA, SA, JA, and ethephon was supplied to the Hewitt medium as described elsewhere ( Van Bockhaven et al, 2015 ).…”
