Characterization of ermB Gene Transposition by Tn 1545 and Tn 917 in Macrolide-Resistant Streptococcus pneumoniae Isolates

Abstract: In Streptococcus pneumoniae, the ermB gene is carried by transposons, such as Tn917 and Tn1545. This study investigated the relationship between macrolide resistance and the presence of the ermB gene on Tn917 or Tn1545 in 84 Japanese pneumococcal isolates. Macrolide-resistant strains were classified into two groups as follows. Group 1 (19 strains) showed a tendency to high resistance to erythromycin (MIC at which 50% of isolates are inhibited, 4 mg/liter; MIC at which 90% of isolates are inhibited [MIC 90 ], … Show more

“…46 For MLSB resistance genes, ermB gene was enriched 126.5-fold (compared with control soil sample), and ermB gene has been found on transposons, such as Tn917 and Tn1545. 47 Plainly, those ARGs which are associated with MGEs can move from one species to another through horizontal gene tranfer mechanisms, 34 with easy dissemination and obstinate persistance in various environments.…”

“…There are some geographical differences in the prevalence and spread of transposons carried erm (B) gene in pneumococci. In Italy and Spain, the most frequently reported transposons in S. pneumoniae are Tn 6002 and Tn 3872 , whereas in Japan Tn 917 was found to be the most common [ 14 , 23 , 24 , 33 ]. Tn 3872 is a composite, mobile element resulting from the insertion of the erm -containing Tn 917 transposon into Tn 916 [ 8 , 23 ].…”

“…The macrolide resistance gene ermB was also found to be abundant in samples across all broiler sheds. Similar to the tet genes, ermB was also detected in a wide range of bacterial species [29] and associated with conjugative transposons [36][37][38], which may explain the wide dissemination of the gene in the environment.…”
