Characterization of Immunological Responses toBorreliaImmunogenic Protein A (BipA), a Species-Specific Antigen for North American Tick-Borne Relapsing Fever

Abstract: Borrelia species transmitted by soft or hard ticks cause tick-borne relapsing fever (TBRF). This is a debilitating disease distributed worldwide but is likely underdiagnosed or misdiagnosed as Lyme disease due to poorly developed diagnostic tests. Borrelia turicatae , Borrelia parkeri , and Borrelia hermsii are three TBRF species previously implicated in human disease in North America.

“…We used Bt-rBipA from B. turicatae because of the vector's distribution in Mexico. 3 Evidence supports that BipA can discriminate between borreliae RF infections in North America, 6 and it is likely that the three patients (cases 1, 2, and 4) were in contact with B. turicatae. Although one patient was suspected by the referring physician of being infected with B. burgdorferi, reactivity to GlpQ indicated exposure to an RF borreliae.…”

“…4 Although GlpQ is highly conserved between species of RF spirochetes and serological cross reactivity occurs regardless of the species causing infection, antibody responses to BipA are likely specific to a given species. 6 This suggests that the patient in case 3 (rGlpQ positive, Bt-rBipA negative) was exposed to another species of RF spirochete. Historical reports identified Ornithodoros talaje, which transmits Borrelia mazzotti, in northern Mexico.…”

“…Region 2 contained ribonucleoside-diphosphate reductase 2 subunit beta (nrdF), ribonucleosidediphosphate reductase 2 subunit alpha (nrdE), a ribonucleotide reductase Class Ib (nrdI), and plasmid partitioning genes (1). A gene encoding the Borrelia immunogenic protein A (bipA), an important diagnostic antigen (24)(25)(26), was found in region 2 of all isolates except for B. anserina BA2, which did not encode a homologue. Intact phagerelated genes were also found in region 2 (e.g.…”

“…A glycerophosphodiester phosphodiesterase (GlpQ) protein ( Schwan et al, 1996 ) can differentiate RF antibodies from Lyme disease (LD) associated antibodies; the gene is known to be highly conserved among RF Borrelia species ( Lopez et al, 2016 ). Additional protein antigens that have been identified which have demonstrated higher levels of species-specificity among RF Borrelia are Borrelia immunogenic protein A (BipA) ( Lopez et al, 2010 , 2013 ; Curtis et al, 2022 ) and Borrelia miyamotoi membrane antigen A (BmaA) ( Harris et al, 2020 ). Here, we synthesized these previously characterized RF recombinant targets in addition to identifying and synthesizing a BipA homolog target in B. miyamotoi .…”
