Characterization of Little millet (Panicum sumatrense) varieties using Morphological descriptors and SSR based DNA fingerprinting

Abstract: Little millet varieties are generally distinguished by morphological descriptors which are being used for seed certification and DUS characterization [1]. But in practical terms, these key differentiation descriptors between varieties of little millet are very fewer and hence difficult to differentiate germplasm accessions. Germplasm registration in NBPGR needs DNA fingerprint to show the uniqueness of germplasm in comparison to existing varieties. DNA fingerprinting is a better option to identify unique marke… Show more

“…These qualitative traits could further be evaluated in other trials and could be used as morphological markers for the identification of true to type landraces from the hilly regions. Similar results for qualitative analysis and DUS characterization were also reported by (Natesan et al, 2020) Genotypes with arched inflorescence, compact panicle and round grains were included in the cluster I and cluster III (Table 9). Among these two clusters, cluster III contained genotypes with pigmentation at the leaf sheath while pigmentation was absent in the genotypes grouped under cluster I.…”

“…As a result of that, unlike rice, wheat, and other cereals, the crop has still not achieved breakthroughs in terms of gene discoveries, genetic improvement, and yield enhancement. Also, due to a lack of enough studies, ‘Omics’ information related to the crop has remained scarce ( Natesan et al., 2020 ).…”

“…The current methods of fingerprint construction include polyacrylamide gel electrophoresis (PAGE), capillary electrophoresis with fluorescence detection, etc. [35][36][37]. With the rapid development of capillary electrophoresis with fluorescent SSR markers to construct DNA fingerprints, the obtained results can be converted into strings, barcodes, and quick response (QR) codes with the help of fingerprints [38,39].…”
